Operation method of microphotography
(1) component
Assembly drawing of Olympus PM- 10AD microphotography system.
When choosing 135 camera to shoot, the 135 camera should be installed on the upper part of the adapter PM-PBS, and the focusing telescope PM-VTM should be installed on the front end. Then add an NFK photographic eyepiece (for example, 3.3x) with an appropriate multiple on the straight tube of the microscope, and install PM-PBS on the eyepiece. Finally, install the connecting wires of PM-PBS and PM-CBAD, and connect the power supply of microscope and PM-CBAD respectively. Pay attention to check whether the voltage selection range matches the power supply used.
(2) Check before use
After turning on the microscope and exposure controller, before installing the film, check whether the equipment works normally according to the following methods. The method is as follows: firstly, pull the optical path control lever on the exposure device PM-PBS out of a position (green ring), and adjust the exposure mode on the exposure controller PM-CBAD to the "AUT0" position, so that the ASA speed reaches the ASA value of the film used, and then check as follows:
1) The "Safety" green indicator on the exposure controller should be lit.
2) When the shutter button is lightly pressed, the "Work" light should be on.
3) When closing the shutter, the "Work" light should be turned off, and at the same time, the "film winding" light can be seen and the motor sound of film winding can be heard.
(3) Installation of film
Generally, the camera is not disassembled, and the film is directly loaded or returned to this equipment. The correct loading method is as follows:
1) Pull the strap button outward and the back cover will open. Put 135 film in the film slot under the rewind button, and then reset the rewind button.
2) Insert the film head into the gap of the reel, and be careful not to let the film head protrude from the other end of the sleeve seam. Press the pause/winding button on the controller to drag the film until all the film holes at both ends of the film are embedded in the film winding gear.
3) Cover the back cover again, and then press the film feed key to drag the film for 2-3 times until "1" is displayed on the counter.
4) In order to avoid forgetting the type of film used, it is best to tear off a corner of the packaging box and insert it into the insertion cavity outside the back cover.
(4) Return the film
When all films have been shot, the "Film End" light on the controller will light up and an alarm will appear.
Ring the bell. Take off the camera or rewind the film directly and press the "Time Off" button to stop the alarm sound.
1) Rotate the eject button on the camera 90 degrees counterclockwise.
2) Pull out the film ejection handle and pour the film into the film box clockwise until the tension suddenly decreases.
When there is no resistance, it means that the film has been completely poured into the film box. At this time, you can open the lid and take out the finished film.
3) After the film is completely returned, the film ejection handle will automatically reset.
(5) Black and white micrograph
First, focus the specimen accurately, and select the specimen area to be photographed with the focusing telescope. The shooting steps are as follows:
1) Set Mode/Exposure Time to Automatic; Press "35" in the "Format" selection key; Adjust "ASA" to the value of the photosensitive speed ASA of the film used; Adjust the "RE-CIPROCITY" to the corresponding value of the film (please refer to the plastic indicator board at the bottom of the controller, which should usually be placed in the "4" position unless otherwise specified).
2) Check whether the safety indicator light is green. If it is red, the following checks should be made: a. If it is red,
The flash and alarm sound at the same time, indicating overexposure. It is generally inappropriate to use neutral density filters to reduce illumination brightness.
Reduce the voltage to reduce the illumination brightness. B. If the red light stays on, it means that the exposure is insufficient, and the exposure should be increased.
Increase the brightness.
3) Adjust "exposure ADJ" to an appropriate value. Generally, when the samples are evenly distributed and the vision is bright, it should be adjusted to (1); If the sample is unevenly distributed or irradiated in other ways, appropriate adjustments should be made:
4) Pull down the exposure button on the controller, and the "Work" light will turn on with the exposure. After the exposure, the "Work" light will be turned off and the film will automatically enter the film position.
(6) 135 color film photomicrography
The basic method of color photography is the same as that of black-and-white microphotography, but the color temperature adjustment is different. The following only introduces the temperature adjustment method;
1) Install the color temperature detector PM-CTR on the left side of the automatic exposure machine PM-PBS.
2) After the specimen is in focus, move the field of vision to a place where there is no specimen.
3) Move the light path control lever to the outermost (yellow belt); Adjust the color temperature to "D" (daylight film) or "T" (lighting film).
4) Add LBT (optical film) or LBD-2 (solar film) filter to the microscope, and adjust the brightness of the microscope to make the indicator light on the color temperature regulator light up to A (yellow); If the green light above a indicates that the color temperature is too high; A The red light below indicates that the color temperature is too low.
5) Push the light path control lever inward to the green ring, and you can enter according to the above black and white film method.
Take micrographs.
(7) Manual microphotography
Under special circumstances, in order to obtain better photographic effect, operators who often need to manually change exposure conditions can also choose to manually take micrographs. These methods are as follows:
1) Use the Mode/Exposure Time button to select the exposure time (from 1 sec to 40 minutes).
2) Press the shutter button, and the shutter will be exposed at the set time.
3) If the exposure time exceeds 40m, turn the "Mode" button to T, then press the shutter button, and the shutter will remain open until the "Time Close/Winding" button is pressed.
(8) Application of automatic exposure locking device
The same exposure is usually used when shooting different positions or different parts of the same specimen.
Time, and then press the "automatic exposure lock" key. In the subsequent shooting, the same exposure time will be maintained, and the "Time Memory" light will flash until the "AE Lock" key is pressed again.
(9) Repeated exposure: the operation is as follows:
A. First, adjust the "ASA" button to 8 times the ASA value of the film (for example, adjust 100ASA film to 800).
B Press the "L" key selected by "Format", and the same negative can be exposed multiple times.
C. Press the "35" key and "Reel" key after completion, and the film will automatically advance one position.
